Catecholamines inhibit insulin-stimulated glucose transport in adipocytes, in the presence of adenosine deaminase.

چکیده
Epinephrine, norepinephrine and isoproterenol completely inhibited insulin-stimulated 2-deoxyglucose uptake by rat adipocytes, but only when adenosine was prevented from accumulating in the incubation medium by addition of adenosine deaminase. Basal uptake rates were not affected. The effects were not observed in the presence of adenosine deaminase plus N6-phenylisopropyl adenosine (a non-metabolizable adenosine analogue), suggesting that it is the lowered adenosine level rather than the presence of the enzyme itself which allows the catecholamines to inhibit insulin action. The inhibitory effects of the catecholamines were blocked by propranolol but not phentolamine, suggesting that they are mediated via beta-adrenergic receptors.

Propionic acid and butyric acid inhibit lipolysis and de novo lipogenesis and increase insulin-stimulated glucose uptake in primary rat adipocytes
Fermentation of dietary fibers by colonic microbiota generates short-chain fatty acids (SCFAs), e.g., propionic acid and butyric acid, which have been described to have "anti-obesity properties" by ameliorating fasting glycaemia, body weight and insulin tolerance in animal models. In the present study, we therefore investigate if propionic acid and butyric acid have effects on lipolysis, de nov...
متن کاملUnderstanding the Mechanism Underlie the Antidiabetic Activity of Oleuropein Using Ex-Vivo Approach
Background: Oleuropein, the main constituent of olive fruit and leaves, has been reported to protect against insulin resistance and diabetes. While many experimental investigations have examined the mechanisms by which oleuropein improves insulin resistance and diabetes, much of these investigations have been carried out in either muscle cell lines or in vivo models two scenarios with many draw...
متن کاملInsulin-stimulated glucose transport and insulin internalization share a common postbinding step in adipocytes.
We recently demonstrated that chymotrypsin substrate analogues inhibit receptor-mediated insulin internalization in isolated rat adipocytes. In this study, the effect on glucose transport of inhibiting insulin internalization with these agents was examined. Glucose transport was assayed by measuring [3H]-2-deoxyglucose uptake, and internalized insulin was measured after rapidly dissociating sur...
متن کاملGLUT4 trafficking in insulin-stimulated rat adipose cells: evidence that heterotrimeric GTP-binding proteins regulate the fusion of docked GLUT4-containing vesicles.
Agents that activate the G-protein G(i) (e.g. adenosine) increase, and agents that activate G(s) [e.g. isoprenaline (isoproterenol)] decrease, steady-state insulin-stimulated glucose transport activity and cell-surface GLUT4 in isolated rat adipose cells without changing plasma membrane GLUT4 content. Here we have further examined the effects of R(s)G(s) and R(i)G(i) ligands (in which R(s) and ...
متن کاملAssociation between Serum Adenosine Deaminase Activity and Blood Glucose Level in Gestational Diabetes
Background &Objectives: Adenosine deaminase (ADA) is a marker of T cell activation. ADA catalyzes the deamination of adenosine and modulates the bioactivity of insulin. The aim of this study was to investigate a possible relationship between ADA activity and various clinical and metabolic parameters, including fasting plasma sugar (FBS) and glycated hemoglobin (HBA1c), in gestational diabetes (...
